Elevated concentrations of leukocytes in semen have been associated with genital tract infection has negative effect on semen quality and IVF and embryo transfer.

 According to the World Health Organization (WHO), the threshold value for a normal seminal white blood cell (WBC) count is 1×106/ml. Peroxidase enzyme in leukocytes raise the reactive oxygen spices (ROS) and defend against pathogenic agents. So polymorphoneuclear can be counted by peroxidase staining. To count leukocytes in semen this assay is simple and accurate

 ROS that produced by WBC could increase the fragmentation in DNA of sperms.

The most common type of WBC in semen are polymorphonuclear or granulocytes (50–60%), followed by macrophages (20–30%) and T-lymphocytes (2–5%).

Reproductive tract infection is an important cause of male infertility, and anti-inflammatory treatment is an important means to improve male fertility.

The increase of leukocytes in semen may be a sign of reproductive system infection. When the concentration of leukocytes is more than 1 × 106/mL, sperm quality will decrease.

Leukocytes had a positive association with normal morphology and progressive motility in semen samples at a concentration of 0-1 × 10(6)/mL. The findings suggest that the association between leukocytes (PMNL) and semen quality might be concentration dependent. More than this range leukocytes negatively impact on semen quality as a result of the presence of reactive oxygen species (ROS) that ROS are harmful to spermatozoa.

Leukocytospermia, defined by the World Health Organization (WHO) as more than 1 million leukocytes per milliliter, has an incidence of 10-20% in the general population and is especially common in infertile men. However, lower concentrations of seminal leukocytes (0-1 × 106/mL) are still more widespread, and are seen even in the absence of infection .

Storage conditions 

LD kit should be kept out of sun and stored at the temperature of 2~8° C for 6 months.

Sperm sample
Semen samples should be collected in a sterile container. The staining sperms should be performed. Fresh sample is needed.
